Asparaginase (EC 3.5.1.1, ASNase) is an amide hydrolase that can remove the amino group from L-asparagine to produce L-aspartic acid and ammonia. This enzyme has anti-tumor activity and is widely used in food and medicine. ASNase catalyzes the hydrolysis of asparagine into aspartic acid and ammonia. Ammonia, in a strong alkaline environment, reacts with hypochlorite and phenol to form the water-soluble dye indigo blue, which has a stable solution color. It has a characteristic absorption peak at 630nm, and by detecting the rate of increase of ammonia, the enzyme activity can be calculated.
