Alanine aminotransferase, formerly known as glutamate pyruvate transaminase, abbreviated as ALT or GPT (EC 2.6.1.2), is widely present in animals, plants, microorganisms, and cultured cells. It catalyzes the transamination reaction between amino acids and keto acids and plays an important role in amino acid metabolism. GPT/ALT catalyzes the transamination reaction between alanine and α-ketoglutarate, producing pyruvate and glutamate; the addition of 2,4-dinitrophenylhydrazine solution not only terminates the above reaction but also reacts with pyruvate to form 2,4-dinitrophenylhydrazone, showing a brown-red color in alkaline solution. The activity of the enzyme can be calculated by reading the absorbance at 520nm.
