ADPG pyrophosphorylase (AGP, EC 2.7.7.27) is an enzyme that plays a key regulatory role in the synthesis of plant starch, catalyzing the reaction of l-phosphoglucopyranose (G-1-P) with adenosine triphosphate (ATP) to form adenosine diphosphate glucose (ADPG), the direct precursor of starch synthesis. In plants, it is mainly found in storage organs and leaves. The reverse reaction catalyzed by AGP produces G1P. In the reaction system, added phosphohexose isomerase and glucose-6-phosphate dehydrogenase successively catalyze the production of 6-phosphogluconate and NADPH. The increase in NADPH at 340nm can be used to calculate AGP activity.
