Adenosine deaminase (ADA, EC 3.5.4.4) is a pyrophosphatase enzyme and a key enzyme in the metabolism of purine nucleosides, which is significantly related to the immune activity of cells in the body. ADA catalyzes the hydrolysis of adenosine to produce inosine and ammonia. Ammonia, in a strong alkaline environment, reacts with hypochlorite and phenol to form the water-soluble dye indigo blue, which has a stable solution color. It has a characteristic absorption peak at 630nm, and by detecting the rate of increase of ammonia, the enzyme activity can be calculated.
