Acetyl-CoA carboxylase (ACC, EC 6.4.1.2) is widely present in the biological world. It catalyzes the carboxylation of acetyl-CoA to form malonyl-CoA in organisms, which is a key enzyme in the synthesis of fatty acids and many secondary metabolic products. The activity of ACC determines the speed of fatty acid synthesis and the level of oil content to a certain extent. ACC catalyzes the generation of malonyl-CoA, inorganic phosphorus, and ADP from acetyl-CoA, NaHCO3, and ATP. ADP and phosphoenolpyruvate are acted upon by pyruvate kinase and lactate dehydrogenase, respectively, causing NADH to be oxidized to NAD+. The decrease in NADH at 340nm is measured to calculate the enzyme activity of ACC.

UV-Vis spectrophotometer, 1mL quartz cuvette (optical path 1cm), desktop centrifuge, pipette, mortar, ice, and distilled water.
