Acetolactate synthase (ALS, EC 2.2.1.6) is a key enzyme in the biosynthesis pathway of branched-chain amino acids, which only exists in plants and microorganisms and is an important target for green herbicides. ALS catalyzes the condensation of two molecules of pyruvate to form acetolactate, which then decouples to form acetoin under the action of sulfuric acid. The product reacts with a color developer to form a colored substance with a characteristic absorption peak at 525nm. By detecting the increase rate of this colored substance, the ALS enzyme activity can be determined. The reaction equation is: 2 pyruvate = 2-acetolactate + CO2.
