6-Phosphogluconate dehydrogenase (G6PDH; EC 1.1.1.49) is widely present in animals, plants, microorganisms, and cultured cells, and is a key enzyme in the pentose phosphate pathway, maintaining the level of NADPH in cells. NADPH, in turn, maintains the level of glutathione in cells, thereby protecting cells from oxidative damage. Therefore, the activity of G6PDH can reflect the biosynthetic and antioxidant capabilities of an organism to a certain extent. G6PDH catalyzes the oxidation of 6-phosphogluconate to 6-phosphogluconolactone, while reducing NADP+ to NADPH. Traditional methods involve detecting the absorbance of NADPH at 340nm. Due to the low molar extinction coefficient (ε) of NADPH, this method has low sensitivity and is highly susceptible to interference. This assay kit provides a simple, sensitive, and rapid detection method: the NADPH produced in this enzymatic process reacts with a specific color-developing agent to form a colored substance with a maximum absorption peak at 450nm. By detecting the increase rate of this colored substance at 450nm, the activity of G6PDH can be calculated.
