Δ1-Pyrroline-5-carboxylate synthase (P5CS) is a key enzyme in proline biosynthesis in plants and also a rate-limiting enzyme in proline synthesis when plants are under stress; proline, as an osmotic regulator, is closely related to plant adaptation to drought and salt stress. Therefore, P5CS plays an important role in regulating the metabolic level of proline. Δ1-Pyrroline-5-carboxylate synthase (P5CS) catalyzes the conversion of glutamate to γ-glutamate semialdehyde while oxidizing NADPH to NADP+. By detecting the decrease rate of NADPH at 340nm, the enzyme activity of P5CS can be determined.
