γ-Glutamyl kinase (γ-GK, EC 2.7.2.11) is one of the key enzymes in the proline biosynthesis pathway. It catalyzes the first step of the reaction from glutamate to proline. This assay kit utilizes γ-GK to catalyze the phosphorylation of glutamate, which is further converted to γ-glutamyl isohemocyanine, and in acidic conditions, forms a Hydroxamate-Fe3+ complex with Fe3+. By detecting the OD value of this complex at 535 nm, the enzyme activity of γ-GK can be determined. The reaction catalyzed by this enzyme is: ATP + L-glutamate = ADP + L-glutamate 5-phosphate.
Additional Materials and Equipments Required
Visible spectrophotometer, 1mL glass cuvette (optical path 1cm), water bath, centrifuge, pipette, mortar, ice, and distilled water.
